Proteinase K is a broad-spectrum serine protease that hydrolyzes various peptide bonds. It works at 20–60°C and pH 7.5–12, tolerates up to 2% SDS or 4 M urea, and remains active in EDTA-containing buffers. Calcium enhances its stability but is not required for activity.
